Riassunto
In the last years, the antibiotic resistance phenomenon has threatened healthcare systems all over the world and only a few newly developed antimicrobial drugs are reaching the pharma market. Among the potential alternative antibacterial treatment, phage therapy emerged as one of the most promising therapeutic options. The present thesis aims to evaluate phage therapy efficacy by performing a systematic review of the clinical case reported during the last twenty years. Data obtained by 38 selected articles were analyzed. Data related to bacterial species, type of infection, phage formulation administered with relative schedule, antibiotic drugs administered before and during the therapy, and outcomes were gathered. The result is promising, with 67% of the patients fully recovered and with bacterial eradication occurring in 90%. In most of the cases, the eradication was obtained when phages were added as an adjuvant to antibiotics, which were not able alone to cure infections. In comparison to clinical trials based on phage therapy reported in the literature, single case reports showed better outcomes. This is due to the viral nature of phages and their specificity and mechanism of action, suggesting that the rules for setting a trial based on phages should be different compared to synthesis molecules such as antibiotics.
