Riassunto (Inglese)
Accreditation according to the UNI CEI EN ISO/IEC 17025:2018 standard attests to the competence of testing laboratories and the reliability of their results, requiring, among other requirements, that the suitability of test methods for their intended use be demonstrated. In the dairy sector, total bacterial count (TBC) represents a parameter of the hygienic quality of milk, the determination of which is required by law. Traditionally quantified using the culture-based method UNI EN ISO 4833-1:2022, it can be determined more rapidly using automated systems such as BacSomatic™. The aim of this study was to validate, at the internal laboratory of a Tuscan dairy plant, an in-house method for the determination of TBC in ovine milk using BacSomatic™.
Preliminary verification of the BacSomatic™ system was performed on ovine milk samples through studies of linearity, carry-over, repeatability, and limit of quantification, in order to assess its proper functioning and performance. Subsequently, the results obtained with BacSomatic™ were compared with those obtained using the reference method UNI EN ISO 4833-1:2022 by means of linear regression, in order to define the conversion relationship, and repeatability, accuracy, and reproducibility were evaluated. Validation of the in-house method was carried out according to the criteria of UNI EN ISO 16297:2020 and ISO 21187:2021. Finally, measurement uncertainty was estimated by considering the main components associated with the analytical result.
The preliminary verification of the BacSomatic™ system showed adequate performance: the limit of quantification was 50.81 IBC/µL, while the mean carry-over was 0.34%, below the acceptance limit, and the instrument showed linearity over the range considered. Instrument repeatability was found to comply with the expected performance. Comparison with the reference culture-based method allowed the conversion relationship to be defined. The method showed good repeatability and accuracy, with z-scores of 1.36 and 1.96 in the interlaboratory proficiency testing scheme, and a reproducibility of 0.043 log₁₀, below the limit of 0.16 specified by ISO 16297:2020. The method was therefore considered suitable for the determination of TBC in raw ovine milk at the dairy's internal laboratory.
