Riassunto (Inglese)
Luminescent complexes based on trivalent lanthanide ions, such as Europium(III), are innovative tools for bioimaging and diagnostics. Thanks to f-f transitions, these systems offer narrow emission bands, large Stokes shifts, high stability, and negligible photobleaching. Since these transitions are forbidden, efficient excitation requires the use of π-conjugated chromophoric ligands (the antenna), whose lipophilicity reduces the water solubility and biocompatibility of the complex.
Glycoconjugation overcomes these limitations by increasing selectivity toward specific targets, such as bacterial lectins. Opportunistic pathogens like Pseudomonas aeruginosa produce lectins (LecA and LecB) essential for cell adhesion and the stabilization of biofilm, a protective matrix highly resistant to antibiotics.
My thesis focuses on the development of novel glycoconjugated Eu(III) complexes designed to target lectins and enable early biofilm detection. Through multi-step synthesis and click chemistry (CuAAC), three new complexes were synthesized featuring a longer flexible PEG spacer compared to previous ones: Eu-PEG-Tz-PEG-β-Gluco, Eu-PEG-Tz-PEG-β-Galacto, and Eu-PEG-Tz-PEG-α-Manno. The extended linker is engineered to promote multivalent interaction with the target, reduce steric hindrance, and increase solubility in aqueous media. The complexes were evaluated using fluorescence assays to test their biofilm-labeling capabilities.
