The present thesis focuses on three main points:
1. To fully characterize the localization of PD-L1 at the plasma membrane level on scales <100 nm by super-resolution immunofluorescence and colocalization with raft-markers such as GPI and caveolin1.
2. To study the diffusive dynamics of PD-L1 at the plasma membrane level using a probe genetically encoded fluorescent protein bound and an imaging scheme, using microscopy of fluorescence, able to quantitatively reveal diffusional dynamics.
3. Verify the existence of regulation of PD-L1 expression in the membrane by transcription factors belonging to the Polycomb Proteins group by flow cytometry, quantitative analysis of immunofluorescence and quantitative PCR and using transcriptional silencing strategies by siRNA.
