Huge improvements in plant productivity are needed to cope with the increasing demand of food, feed and other plant-derived products in the coming decades. Fully exploiting the photosynthetic potential holds great promises for biomass increases in our major crops, but the employment of such potential in the breeding programs relies on the identification of the genetic variation available in nature. In Arabidopsis thaliana, the use of cytoplasmatic hybrids (also termed cybrids) allowed to identify two chloroplast encoded missense variations potentially responsible for an improved performance of PSII and NPQ. In this study, transcriptional and functional analyses on the allelic variants of the matK (ATCG00040) and ndhG (ATCG01080) genes were performed. Both the genes appeared to be expressed and, in case of the ndhG, NDH complex activity monitoring revealed that the mutation seems not to be not responsible for a functional impairment of the whole NDH complex. In case of the matK gene, no functional analysis was performed and no data is available on whether the encoded protein retains its function. As the genetic transformation of Arabidopsis – final goal of this study – could not be achieved, extensive description of the troubleshooting process during the expression vector construction is also provided.
