ABSTRACT
Seeds of L. mutabilis and L. albus were incubated in various osmotic solutions of NaCl and mannitol in order to determine the relative importance of ionic toxicity and osmotic component of salt stress on germination. The seeds were sowed in the absence (control) or in the presence of NaCl or mannitol 100, 200 and 400 mM. Germination and root length related data were collected for the consecutive eight days after sowing. In addition, at day four and eight we measured: cotyledon weight, root weight and root diameter. The biometric parameters showed several differences on the germination strategy between the two species and, between different treatments on the same species. L. albus showed reduced stress effects from the osmotic pressure given by the mannitol. At the contrary, L. mutabilis is less affected from high sodium chloride concentration and it seems to be able to control better the stress, if compared with L. albus, without showing specific water uptake adjustments. At the valuation of the seeds water content, there were no evident differences between the species. The different tolerance mechanisms to salt between the two species in relation to its exclusion or compartmentalization, was studied thanks to atomic absorption spectrometry of ionic content in the different samples. In addition, we analyzed the total sugars amount within the cotyledon of both species. The results showed that at “time zero” (dry seed), L. mutabilis contains an higher sugars level but, differently from L. albus, seems to not use it as response to salinity stress. Finally, we analysed the cotyledonal proteins using SDS-PAGE technique that showed a particular pattern expressed by L. mutabilis cotyledons treated with low NaCl concentration. It is an evidence that a different metabolism between the two species and between different treatments is activated.

RIASSUNTO
Semi di L. mutabilis e L. albus sono stati incubati in soluzioni osmotiche di NaCl e mannitolo, al fine di valutare l’impatto della tossicità ionica e della componente osmotica dello stress salino in fase germinativa. I semi sono stati fatti germinare in assenza (controlli) o presenza di NaCl o di mannitolo 100, 200 e 400 mM. Per gli otto giorni seguenti la semina, sono stati raccolti i dati relativi a germinazione e lunghezza della radice. A quattro ed otto giorni sono stati inoltre misurati: peso cotiledone, peso radice e diametro radicale. I parametri biometrici hanno evidenziato numerose discordanze nelle strategie di germinazione tra le due specie e, all’interno delle singole specie, tra i trattamenti. La pressione osmotica è sopportata meglio da L. albus, che in presenza di solo mannitolo mostra minori segni di stress. L. mutabilis invece risente meno di alte concentrazioni di cloruro di sodio e sembra essere in grado di controllare meglio il fenomeno stressante, rispetto a quanto faccia L. albus, senza mostrare particolari adattamenti legati all’uptake idrico. La valutazione della componente idrica nei semi non ha però evidenziato particolari differenze tra le due specie. Il diverso meccanismo di tolleranza al sale tra le due specie in relazione alla esclusione e compartimentalizzazione è stato approfondito attraverso l’analisi tramite spettrometria ad assorbimento atomico del contenuto ionico dei diversi campioni. Sui cotiledoni di entrambe le specie è stata poi condotta l’analisi degli zuccheri liberi. I risultati mostrano che al tempo zero (seme non imbibito) L. mutabilis può contare su un quantitativo di zuccheri maggiori ma che, a differenza di L. albus, che sembra non utilizzare per rispondere allo stress salino. Il lavoro si è concluso con l’analisi delle proteine tramite SDS-PAGE che ha evidenziato un pattern molto particolare espresso nei cotiledoni di L. mutabilis sottoposti a trattamento con 100 mM di NaCl. Risulta infatti evidente come venga attivato un metabolismo diverso tra le due specie, e fra i diversi trattamenti.
