Riassunto analitico : La mancata o alterata espressione dell’enzima PTEN comporta un’aumento della formazione di AKT, favorendo la proliferazione e sopravvivenza cellulare, il potenziale metastatico e la neoangiogenesi, mentre l’alterata o mancata espressione delle E-caderine causa un difetto strutturale delle giunzioni di adesione intercellulari, provocando la transizione epitelio-mesenchimale, con aumento dell’invasività e resistenza all’apoptosi. Lo scopo della tesi è stato quello di investigare l’espressione di PTEN e delle E-caderine in relazione ai diversi istotipi di neoplasie mammarie maligne canine (CMMT), in modo da verificare un’eventuale correlazione tra l’espressione delle due proteine e le caratteristiche istopatologiche e cliniche di malignità. Quaranta CMMT (15 complessi e 25 semplici) sono stati sottoposti ad esame immunoistochimico per valutare l’espressione di PTEN e delle E-caderine. Questo studio conferma il l’associazione tra modificazioni della via PTEN/AKT e dell’espressione delle E-caderine con una maggiore aggressività della neoplasia, testimoniata da neoplasie semplici, più aggressive, e ridotta sopravvivenza. I risultati dimostrano, infatti, come PTEN sia correlato ad un minor indice proliferativo, ad una maggior sopravvivenza e ad una corretta espressione delle E-caderine; esso infatti è la proteina regolatrici più importante di questa via e la sua riduzione è associata al peggioramento della patologia. Una corretta espressione delle E-caderine è stata rilevata nelle forme con istotipo complesso, meno aggressivo. I risultati delle indagini confermano come la vlautazine di questi marker possa fornire importanti informazioni circa la prognosi delle neoplasia mammarie canine, in quanto la contemporanea mancata espressione di queste proteine è correlata alle caratteristiche clinico-istopatologiche di malignità ed a una ridotta sopravvivenza.
Abstract : The lack or the reduced PTEN expression leads to an increase of AKT levels, promoting cellular proliferation and survival, metastatic potential and neoangiogenesis; while the altered expression of E-cadherin leads to a structural defect of cellular adhesion, provoking epithelial-mesenchymal transition with invasiveness increase and apoptosis resistance. The aim of the study is to evaluate the PTEN and E-cadherin expression in malignant canine mammary tumors (CMMT), in order to verify a possible statistic correlation between the expression of the two proteins and histopathologic and clinical features of the neoplasms. Fourty CMMT were submitted to an immunohistochemistry examination in order to evaluate the expression of the proteins. This study confirms the association between modifications of PTEN/AKT pathway and E-cadherin’s expression with a greater malignancy of neoplasias, highlithed by a reduced overall survival and the presence of simple and more aggressive tumors.
Furthermore, the results shows how PTEN is related to a lower proliferative index, to a survival increase and to a correct E-cadherin expression. PTEN, infact, is one of the most important regulator protein of this pathway and its decrease is associated to the disease worsening. A correct E-cadherin expression was mainly detected in the less aggressive complex carcinomas. The results shows how the marker assessment could affect important informations about the prognosis of CMT, since the contemporary lack of the proteins is related to clinc-pathological features of malignancy and reduced overall survival.
