Summary
L’aldoso reduttasi (AR, AKR1B1) è un enzima, NADPH dipendente, appartenente alla famiglia delle aldo-cheto-reduttasi citoplasmatiche. Tale enzima fa parte della così detta via dei polioli nella quale il glucosio viene dapprima ridotto a sorbitolo dall’aldoso reduttasi e successivamente ossidato a fruttosio mediante una reazione NAD+ dipendente catalizzata dall’enzima sorbitolo deidrogenasi. In condizioni di iperglicemia, nelle cellule dove l’uptake di glucosio è indipendente dall’insulina, l’aldoso reduttasi, operando la conversione del glucosio in sorbitolo, porta ad un accumulo di sorbitolo a livello intracellulare che causa alterazioni dell’osmolarità e stress ossidativo con conseguente produzione di specie reattive dell’ossigeno (ROS) in grado di scatenare processi ischemici ed infiammatori responsabili delle complicazioni della patologia diabetica. L’obiettivo della tesi ha riguardato la sintesi e caratterizzazione di nuovi derivati sia poliidrossipiperidinici (azapiranosi) e sia a struttura ossi-immino saccaridica, quali potenziali inibitori dell’enzima Aldoso Reduttasi.
Aldose reductase (AR, AKR1B1) is an enzyme, dependent on NADPH, belonging to the cytoplasmic aldo-keto-reductase family. This enzyme is part of the so-called polyol pathway in which glucose was reduce to sorbitol by the aldose reductase and subsequently was oxidize by the enzyme sorbitol dehydrogenase into fructose. In hyperglycaemia conditions, in cells where glucose uptake is independent of insulin, the aldose reductase, by converting glucose into sorbitol, leads to an accumulation of sorbitol at the intracellular level that causes alterations in osmolarity and oxidative stress. The production of reactive oxygen species (ROS) causes ischemic and inflammatory processes responsible for complications of diabetic pathology. The aim of the thesis was the synthesis and characterization of new azapiranose derivatives and of saccharidic oxy-iminic structure, as potential inhibitors of the enzyme aldose reductase.
