Riassunto
A newly and deathly emerging pathogen is appeared for the first time in 1994 in Australia. The serious damages to livestock and the potential dangerousness for the human population have attracted the
attention of Australian government and public opinion. The Australian government claims new strategies for an early detection of the virus.
In this context, this manuscript presents for the first time a biosensor developed to detect that virus. The strategy chosen to detect the virus is its co-localization through antibodies-coupled functional nanoparticles. A new protocol for protein conjugation and purification of the functional nanoparticles has been developed. Each step of the protocol has been characterized, choosing the suitable techniques among electron microscopy, fluorescence spectroscopy, infrared spectroscopy and ultraviolet-visible spectroscopy. The developed strategy has been compared to the actual virus detection method. The system’s virus response is characterized by many correlated variables. These variables have been optimized with a classic approach (one variable at time), but also a chemometric approach is presented.
The possibilities offered by this strategy represent a proof-of-concept for the development of a fast and portable biosensor.
