RIASSUNTO
Negli ultimi anni sono state compiute diverse ricerche volte ad evidenziare un legame tra patrimonio genetico e comportamento. Molti dei tratti comportamentali hanno mostrato un’alta ereditabilità, ad indicazione del fatto che il profilo genetico esercita un’influenza non trascurabile sul comportamento. Ad esempio, specifiche varianti genetiche sono state associate ad un comportamento pro-sociale od antisociale, così come al comportamento impulsivo, alla capacità di gestire lo stress ed a molti altri tratti.
In questo lavoro ci siamo proposti di indagare quali varianti genetiche giochino un ruolo nei meccanismi decisionali che sono alla base del giudizio morale. A questo scopo, abbiamo selezionato, sulla base dei dati presenti in letteratura, alcuni geni e polimorfismi associati a tratti comportamentali quali l’empatia e l’altruismo, che rappresentano degli endofenotipi che intervengono nelle scelte morali. In particolare abbiamo individuato 4 polimorfismi su 3 geni candidati: la VNTR (Variable Number of Tandem Repeat) DRD4-1/11 e gli SNP (Single Nucleotide Polymorphism) COMT rs4680, OXTR rs53576 ed OXTR rs2268498. Quest’ultimo SNP risulta già associato in letteratura al giudizio morale, ma non ne è mai stata valutata un’eventuale correlazione con altre varianti.
Da un punto di vista sperimentale è stato raccolto il DNA mediante prelievo di saliva a 200 volontari sani, ai quali è stato chiesto di rispondere ad una serie di dilemmi morali proposti da un gruppo di psicologi.
Il mio lavoro di tesi è consistito nella determinazione del genotipo dei 200 campioni per ciascuna delle suddette varianti genetiche. L’analisi del genotipo è stata effettuata utilizzando tre diverse tecniche di biologia molecolare: la PCR (Polymerase Chain Reaction) seguita da elettroforesi su gel di agarosio nel caso della VNTR, la High Resolution Melting (HRM)-PCR per lo SNP rs4680 e la PCR-RFLP (Restriction Fragment Length Polymorphism) per gli altri due SNP. I casi dubbi sono stati risolti mediante sequenziamento secondo il metodo di Sanger.
Dai risultati è emerso che i polimorfismi OXTR rs2268498, COMT rs4680 e DRD4-1/11 in interazione con i dilemmi morali in cui l’aspettativa di vita può essere ridotta o normale, DRD4-1/11 in interazione con COMT rs4680, oltre ad OXTR rs53576, sono in grado di condizionare le risposte ai dilemmi morali che propongono una scelta risolutiva di tipo utilitaristico.


ABSTRACT
In the last years several studies have been conducted to find a link between human genetic heritage and behavior. Most human behavioral traits show high hereditability, indicating that the genetic background exerts a considerable influence on behavior. For example, specific genetic variants have been associated to pro-social and anti-social behavior, as well as to impulsive behavior, ability to manage stress and many other traits.
Aim of this thesis was to investigate which genetic variants could play a role in the decision-making mechanisms underlying moral judgment. To this purpose, based on literature data, we selected a group of polymorphisms associated with behavioral traits such as empathy and altruism, which are endophenotypes involved in moral choices. In particular, we identified 4 polymorphisms on 3 candidate genes: the VNTR (Variable Number of Tandem Repeat) DRD4-1/11, and SNPs (Single Nucleotide Polymorphism) COMT rs4680, OXTR rs53576 and OXTR rs2268498. This SNP has been already associated to moral judgment, but the possible correlation with other variants has not been evaluated yet.
From an experimental point of view, DNA was collected by taking saliva samples from 200 healthy volunteers who were asked to answer a series of moral dilemmas proposed by a team of psychologists.
My experimental thesis work was to genotype 200 samples for the selected genetic variants. Genotype analysis has been performed using different molecular biology techniques, such as PCR (Polymerase Chain Reaction) followed by electrophoresis on agarose gel in the case of VNTR, the High Resolution Melting (HRM)-PCR for the SNP rs4680 and PCR-RFLP (Restriction Fragment Length Polymorphism) for the two other SNPs. The doubtful cases were solved by sequencing according to Sanger’s method.
Our results provide evidence that the polymorphisms OXTR rs2268498, COMT rs4680 and DRD4-1/11 in interaction with moral dilemmas where life expectancy can be reduced or normal, DRD4-1/11 in interaction with COMT rs4680, as well as OXTR rs53576, are able to affect the answers to moral dilemmas offering an utilitarian resolution.
