L’epatite E è una patologia virale a trasmissione oro-fecale il cui agente eziologico Hepatitis E virus (HEV) è un virus appartenente al genere Hepevirus della famiglia Hepeviridae. Il virione è privo di envelope ed il genoma è costituito da RNA a singolo filamento con polarità positiva. Finora sono stati riconosciuti quattro principali genotipi; i genotipi 1 e 2, esclusivamente umani, sono stati riconosciuti come agenti eziologici dei focolai epidemici trasmessi a seguito dell’utilizzo e consumo di acque contaminate nei Paesi in via di sviluppo, mentre i genotipi 3 e 4, sono agenti zoonosici in grado di infettare l’uomo, il suino ed altre specie animali. Tecniche molecolari per l'identificazione del genoma virale hanno contribuito ad aumentare il tasso di rilevamento di HEV in suini, ma a tutt’oggi, la breve durata della viremia e la scarsa resistenza del virus nelle feci rappresentano un importante limite per la diagnosi diretta dell’infezione. La sierologia al contrario può costituire un buon modello diagnostico ed epidemiologico dal momento che anticorpi anti-HEV specifici sono rilevabili anche a distanza di anni dall’esposizione al virus. Nel presente studio, la glicoproteina capsidica ORF-2 di HEV appartenente al genotipo 3 di suino, precedentemente clonata in un sistema di baculovirus ricombinante, è stata espressa in cellule di insetto, purificata e successivamente testata per la messa a punto di una metodica ELISA indiretta per la titolazione anticorpale su sieri di suino. Lo scopo di questa tesi è stato di standardizzare sia le metodiche per la purificazione della proteina che quelle relative al suo utilizzo come antigene in test ELISA indiretti usando sieri suini. I risultati ottenuti hanno confermato le potenzialità dell'antigene purificato per la messa a punto di un test ELISA efficace e applicabile nel controllo degli allevamenti e per il monitoraggio epidemiologico delle specie domestiche e selvatiche.


Hepatitis E is a viral infectious disease mainly transmitted by oro-fecal route. The causative agent, Hepatitis E virus (HEV), is a non-enveloped, single-stranded, positive-sense RNA virus classified in the family Hepeviridae, genus Hepevirus and it consists of four major genotypes. Genotypes 1 and 2, restricted to humans, are responsible for the onset of large outbreaks and epidemics in developing countries through the use and consumption of contaminated water, whereas genotypes 3 and 4, which infect either humans, pigs or other animal species, are responsible for sporadic cases of hepatitis E in both developing and industrialized countries. Molecular techniques for the identification of the viral genome have enhanced the detection rate of HEV in pigs, but to date, the short duration of viremia and the poor resistance of the virus in feces are an important limitation for the direct detection of the infection. On the contrary, serology may constitute a good diagnostic and epidemiological model since anti-HEV specific antibodies persist for years after the infection. In this study a recombinant baculovirus previously generated, encoding for HEV capsid protein ORF-2 belonging to genotype 3 isolated from swine, has been expressed in insect cells, purified and used as antigen in indirect ELISA tests. The aim of this thesis was to standardize the procedures for the protein purification and for its use as antigen for the setting of an indirect ELISA test for the detection of anti-HEV antibodies in swine sera. The results confirmed the potentialities of the purified protein in the development of an ELISA applicable and effective in the control of the farms and for the Epidemiological Monitoring of domestic and wild species.
