Il primo obbiettivo della presente tesi consiste nel testare la resistenza di 16 sierovarianti di Leptospira spp. verso l’antibiotico tigeciclina, con diluizioni a partire da una concentrazione di 128 μg/ml fino a 0,25 μg/ml, utilizzando come controllo la tetraciclina, l’antibiotico maggiormente utilizzato per il trattamento di leptospirosi. Come secondo obbiettivo è stata testata la suscettibilità delle stesse sierovarianti al sale, partendo da una massima concentrazione di 20% di NaCl fino allo 0,0390625%. Per ogni sierovariante è stata allestita una piastra da microtitolazione nella quale è stata saggiata la Minima Concentrazione Inibente (MIC) per la tigeciclina e la soluzione salina. Successivamente tramite colorazione con resazurina è stato possibile leggere i valori di MIC e conseguentemente valutare la Minima Concentrazione Battericida (MBC). Per quanto riguarda la tigeciclina, i valori di MIC ottenuti sono stati tra 4 e 32 μg/ml, mentre quelli di MBC tra 16 e >128 μg/ml. Patoc è risultata essere la sierovariante meno resistente con valori di MIC e MBC di 4 e 16 μg/ml, rispettivamente. Le sierovarianti più resistenti sono risultate Bataviae (32 μg/ml di MIC e 64 μg/ml di MBC) Bratislava (8 μg/ml e 128 μg/ml) e Tarassovi (8 μg/ml e >128 μg/ml). I valori di MIC per quanto riguarda la prova del sale sono risultati essere in un range tra 0.3125 e 10 % di NaCl, mentre i valori di MBC risultano essere tra 0,625 e >20 %. La sierovariante più sensibile è risultata Icterohaemorrhagiae con valore di MIC di 0,3125 %, mentre il valore di MBC corrisponde a 0,625 %. La sierovariante di Leptospira che risulta essere più tollerante a soluzione salina è stata Tarassovi con valore di MIC e MBC di 10 e >20 %, rispettivamente. Per quanto riguarda Pomona e Bratislava, hanno riportato bassi valori di MIC e alti valori di MBC: per Pomona i rispettivi valori di MIC e MBC sono risultati di 1.25 e >20, mentre per Bratislava di 0.625 e 20%. La sierovariante Pyrogenes ha mostrato valori di MIC e MBC nei confronti del sale inferiori a quelli saggiati. Dai risultati ottenuti dalla prova del sale si deduce che molti ceppi sono tolleranti alle diverse percentuali di sale ma essendo ancora pochi gli studi fatti sulla sensibilità al sale di Leptospira spp., per cui ulteriori ricerche dovranno essere effettuate.

The first objective of this thesis is to test the resistance of 16 serovar of Leptospira spp. to tigecycline with different dilutions ranging from 128 μg/ml to 0.25 μg/ml, using tetracycline as a control. The second objective consists to test the susceptibility to a saline solution of the same serovars, starting from concentration of 20% NaCl to a concentration of 0.0390625% NaCl. Microdilution method was used, to evaluate tigecycline and NaCl Minimum Inhibitory Concentration (MIC) of each Leptospira serovar. After the MIC value was determined, by using resazurin the Minimum Bactericidal Concentration (MBC) was evaluated. The results for the tigecycline MIC are between 4 and 32 μg/ml, the results for the MBC are between 16 and >128 μg/ml. Patoc was the least resistant serovar with MIC and MBC values of 4 and 16 μg/ml, respectively. The most resistant serovars were Bataviae (32 μg/ml of MIC and 64 μg/ml of MBC), Bratislava (8 μg / ml and 128 μg / ml) and Tarassovi (8 μg/ml and > 128 μg/ml). The MIC values for the NaCl test were in a range between 0.3125 and 10% of NaCl, while the MBC values were between 0.625 and > 20%. The most susceptible serovar was Icterohaemorrhagiae with a MIC value of 0.3125%, while the MBC value was 0.625%. More tolerant to saline solution Leptospira serovar was Tarassovi with MIC and MBC values of 10 and > 20%, respectively. Pomona and Bratislava showed low MIC values and high MBC values: for Pomona the MIC was 1,25 % and MBC > 20%, while Bratislava MIC and MBC resulted 0.625% and 20%, respectively. The serovar Pyrogenes never survived to all salt concentration tested. Obtained results from the saline solution test showed that many strains are tolerant to different salt percentages. Few studies were carried out on salt susceptibility of Leptospira spp., therefore more researches should be done.
